So the room is on the second floor of the house. It has one double window in it.

The plan is to do two exhaust vents in the ceiling to outside. I will also have two carbon filters running during and after smoking a cigar in the room.

I am going to run a large leather couch in the room with a poker table and one recliner.

Any ideas? What's the best way to seal the backside of the door that comes in and out?

If you have lots of air being pulled from the room, I dont think you want to seal the door. Fresh air will need to come in from somewhere.
No question, this. You have to replace the air being pulled out or your exhaust fans wont run to their potential. Not to mention creating a negative pressure in a room will cause you to start to suck air from anywhere it can get through, which isnt good either.

May as well smoke outside if you can't keep the room heated or cooled. I think I'm going to wait until I can afford a large enough HEPA and charcoal filter system (Rabbit Air?).

That's not enough either in a room connected to other rooms.

What happened here? I'm looking for ideas for the room, some people have given pertinent information which I appreciate.

I'm going to have two exhaust fans in the ceiling and a outdoor air return to equalize and create a negative pressure situation.

The room is heated and cooled, it's in my house

Simple thin film of door weather stripping. Home depot, get the thinest you can for an inside door, 1/8th of an inch should do it im guessing. Will be hard to close the door at first, but it will compress and make a very good seal for years.

I was thinking something like that. I think it's the best way to prevent it from leaking out

And what about the giant 1/2" to 3/4 giant gap at floor level?
